t-Boc-N-amido-PEG6-Tos is a PROTAC linker based on hexaethylene glycol (PEG6) with the molecular formula C₂₄H₄₁NO₁₀S and a molecular weight of 535.65. Its structure consists of three parts: one end is an amino group protected by tert-butyloxycarbonyl (t-Boc), providing removable protection under mild acidic conditions; the middle section is a flexible PEG chain composed of six ethylene glycol repeating units, which enhances water solubility and biocompatibility; the other end is a tosylate (Tos) group, acting as a highly reactive leaving group that undergoes nucleophilic substitution reactions with molecules containing amino or hydroxyl groups.
The production process employs a stepwise synthesis method: first, the PEG6 chain is prepared through ethylene glycol polymerization. Subsequently, an amino group is introduced at the chain end and protected with t-Boc. Finally, the Tos group is linked via sulfonation. This product is widely used in the construction of PROTAC molecules, serving as a key bridging component to connect E3 ubiquitin ligase ligands with target protein ligands.
